The concentration of ammonium was found to be a critical factor influencing callus development. The optimal NH₄⁺ concentration was determined to be 7.0 mM, which is equivalent to 3.5 mM of (NH₄)₂SO₄. Higher ammonium levels severely inhibited both growth and differentiation of rice pollen callus. In contrast, neither KNO₃ concentration nor other medium components had a significant effect on callus development. Chu's N6 medium was specifically designed to enhance the formation, growth, and differentiation of pollen-derived callus in rice.
